Gastric proton pump expression in teleost fishes: nongastric sites of expression in gastric versus agastric fishes

A bomba de protões gástrica ou H/K-ATPase é responsável pela acidificação do estômago nos vertebrados mas pode também ser encontrada em órgãos não gástricos onde é possível que tenha função de ionoregulação. A H/K-ATPase é um heterodímero composto por duas subunidades, e, codificadas pelos genes ATP4A e ATP4B, respectivamente. O gene ATP4A foi identificado no estômago de alguns teleósteos e nas brânquias de um elasmobrânquio mas não existem dados relativos à sua expressão em órgão extra gástricos em peixes teleósteos. Além disto, existem alguns teleósteos (ciprinídeos, peixes-balão) que são agástricos o que levanta a questão sobre se a expressão extra gástrica é dependente da expressão gástrica. Usando o RT-PCR para o ATP4A e através de anticorpos não homólogos para ATP4A e ATP4B, tentei determinar se a H/K-ATPase também é expressa nas brânquias de peixes teleósteos e se esta expressão é dependente da presença de um estômago funcional. Foram examinadas as seguintes espécies: GÁSTRICAS:Oncorhynchus mykiss e Gasterosteus aculeatus. AGÁSTRICAS:Tetraodon nigroviridis, Cyprinus carpio , Misgurnus anguillicaudatus e Daniorerio. Nas espécies gástricas, a H/K-ATPase foi detectada no estômago e na brânquia. Nas espécies agástricas, as análises insilico em bases de dados de genomas e o RT-PC R da brânquia e intestino falharam na identificação de ATP4A e os resultados de “immunoblotting” relativos à subunidade foram maioritariamente negativos. No entanto, as análises de “immunoblotting” da subunidade indicaram a presença de bandas immunoreactivas na brânquia (80-150kDa) e no intestino (110kDa) de algumas espécies. Nos peixes gástricos a subunidade é de aproximadamente 100kDa. A partir destes resultados, pode concluir-se que os teleósteos gástricos exprimem a H/K-ATPase na brânquia e os resultados imunológicos relativos a subunidade da H/K-ATPase nos peixes sem estômago sugerem mais estudos.
The gastric proton pump or H/-ATPase is responsible for stomach luminal acidification in vertebrates but has also been found in non-gastric or gans where it possibly has a role in ionoregulation. H/K-ATPase is a heterodimer composed of an and subunits encoded by the genes ATP4A and ATP4B, respectively. ATP4A has been identified in the stomach of a few teleosts and the gills of an elasmo branch but no data on non-gastric sites of expression exists in teleost fishes. In addition there are some teleosts (e.g. cyprinids, pufferfish) that are also agastric (stomachless) which raises the question of whether extragastric expression is dependent on gastric expression. Using RT-PCR for ATP4A and non-homologous antibodies to ATP4A and ATP4B I set out to determine if H/K-ATPase is indeed also expressed in the gills of teleost fishes and whether this expression is dependent on the presence of a functional stomach. The following species were examined: GASTRIC: Oncorhynchus mykiss and Gasterosteus aculeatus. AGASTRIC:Tetraodon nigroviridis, Cyprinus carpio, Misgurnus anguillicaudatus, Daniorerio. In gastric species, branchial and gastric H/K-ATPase was detected. In agastric species,in silico analysis of genome databases and RT-PCR of gill and intestine failed to identify ATP4A and -subunit immunoblotting results were mostly negative. However, immunoblotting analysis of -subunit indicates the presence of immunoreactive bands in gill (80-150kDa) and intestine (110kDa) of some but not all species. In gastric fishes -subunit is ~100kDa. From these results I can conclude that gastric teleost fishes also express H/K-ATPase branchially and that there is at least an immunologically related subunit of H/K-ATPase in agastric fishes which awaits further characterization.
