Utilização de marcadores na determinação da digestibilidade e tempo de retenção em tartarugas Trachemys scripta elegans (Wied, 1839)

A Parte I deste trabalho aborda o estado da arte, tendo em conta as características das tartarugas em geral e, mais especificamente, da espécie em estudo, a tartaruga Trachemys scripta elegans. Foram também revistas as principais metodologias para determinação da digestibilidade e da taxa de passagem e tempo de retenção, e suas vantagens e desvantagens. Na Parte II apresentamos a componente prática deste estudo, que é constituída por dois trabalhos experimentais (Capítulo 4 e 5). No Capítulo 4 foi determinado o tempo de retenção para tartarugas Trachemys scripta elegans utilizando dois marcadores externos diferentes, o óxido de crómio e a LIPE® (Lenhina de eucalyptus grandis isolada, purificada e enriquecida). Os resultados obtidos descartam a LIPE® como marcador para estes animais devido à elevada taxa de diluição do marcador na água. A utilização do crómio, com uma taxa de recuperação elevada, permitiu verificar que em 12 dias, sete dos nove animais tinham excretado pelo menos 75% do marcador. O pico de excreção do marcador foi atingido até às 144 horas (seis dias) por todos os animais. Em simultâneo foi estimada a produção fecal ao longo do dia, e verificámos que esta foi quase nula durante a noite e foi elevada aquando as horas de alimentação. No Capítulo 5 foi comparada a estimativa da digestibilidade medida pelos marcadores óxido de crómio e cinza insolúvel em ácido com a colheita total de fezes. O óxido de crómio estimou correctamente a digestibilidade da MS, MO, PB e EB quando comparado com a colheita total, enquanto a cinza insolúvel em ácido subestimou a digestibilidade da MS e sobrestimou a digestibilidade da MO, PB e EB quando comparada com a colheita total.
Part I of this work approaches the state of the art, taking into account the characteristics of turtles in general and, more specifically, the ones of this species, the turtle Trachemys scripta elegans. We also reviewed the main methods for determining the digestibility, passage rate and retention time, and their advantages and disadvantages. In Part II we present the practical component of this study, which consists of two experimental studies (Chapters 4 and 5). In Chapter 4 we determined the retention time for turtles Trachemys scripta elegans using two different external markers, the chromium oxide and LIPE ® (Enriched and purified isolated lignin of Eucalyptus grandis). The results ruled out the LIPE ® as a marker for these animals due to the high rate of dilution of the marker in the water. The use of chromium, with a high recovery rate, showed that in 12 days, seven of nine animals had excreted at least 75% of the marker. The peak excretion of the marker was achieved up to 144 hours (six days) for all animals. At the same time fecal output during the day was estimated, and we found that it was almost zero during the night and was high during the hours of feeding. In Chapter 5 we compared the digestibility estimated by two markers, chromium oxide and acid insoluble ash, with total fecal collection. The chromium oxide correctly estimated the digestibility of DM, OM and CE compared with the total collection, while the acid insoluble ash underestimated the digestibility of DM and over-estimated the digestibility of OM and CE when compared to total collection method.
