Análise proteómica de uma estirpe clínica de Staphylococcus aureus ST398 resistente à meticilina

Actualmente a proteómica é uma ferramenta muito utilizada na análise das
diferenças na expressão de genes em estirpes bacterianas. Há muitos anos que
Staphylococcus aureus tem sido reconhecido como um importante agente patogénico
responsável por doenças humanas. A dificuldade no tratamento e o surgimento constante
de múltiplas resistências a antibióticos têem feito deste organismo um importante foco de
estudo.
Para investigar este patógeno foram determinados, através de técnicas de
electroforese bidimensional e técnicas de cromatografia líquida acoplada à espectrometria
de massa, três proteomas diferentes de uma estirpe clínica de S. aureus resistente à
meticilina (MRSA). Recorrendo à técnica 2-DE para obtenção do proteoma
citoplasmático, foram excisados um total de 105 spots, nos quais, por correlação com as
bases de dados de bioinformática, permitiram a identificação de 227 proteínas. No
proteoma da superfície celular exposta foram identificadas 236 proteínas, assim como no
proteoma extracelular foi possível identificar 99 proteínas. Identificaram-se proteínas
relacionadas com funções-base da célula, mas também proteínas relacionadas com
virulência e patogenicidade, tais como a catalase e a proteína isdA, principal responsável
pela infecção em S. aureus, e proteínas relacionadas com a resistência aos antibióticos
como as proteínas de membrana PBP. As duas classes com mais proteínas identificadas
foram a glicólise, no proteoma intracelular, e a tradução nos outros proteomas da
membrana celular exposta e extracelular, reflectindo assim um metabolismo activo.
Estes resultados revelam a importância da proteómica no desenvolvimento do
conhecimento da expressão proteica da estirpe de MRSA, microrganismo este com
diversos e importantes mecanismos de resistência e patogenicidade. Este mapa
proteómico global permite uma melhor compreensão deste agente patogénico, fornecendo
assim novos dados para bancos de dados de proteínas.
Proteomics is presently a powerful tool to analyze the differences in gene expression in bacterial strains. For many years Staphylococcus aureus has been recognized as an important pathogen and responsible for human diseases. The difficulty in treatment and constant new appearance of multiple antibiotic resistance has made this organism an important focus of study. To investigate this pathogen three different proteomes of a methicillinresistant clinical strains S. aureus (MRSA) were determined by techniques of twodimensional electrophoresis and liquid chromatography-mass spectrometry. Using 2-DE for cellular proteome were excised 105 spots, in which, for correlation with bioinformatics databases, we identified 227 proteins. Considering the exposed cell surface proteome and the exoproteome, 236 and 99 proteins were identified respectively. Proteins related to basic cell functions were found, but also proteins related to virulence and pathogenicity, like catalase and isdA, main responsible for S. aureus infection, and proteins related to antibiotic resistance membrane proteins PBP. The two classes with more proteins identified were glycolysis in the intracellular proteome and translation in the othe exposed cell membrane and exoproteome, this reflects an active metabolism. These results highlight the importance of proteomics to development knowledge of protein expression of MRSA, this microorganism with several important mechanisms of resistance and pathogenicity. This global proteomics allows a better understanding of this pathogen, also providing new data for proteins databases.
