Caracterização Físico-Química e Microbiológica de Águas Residuais de Lagares de Azeite. Selecção de Leveduras para Aplicação ao Tratamento de Águas Ruças

Portugal é um dos dez maiores produtores mundiais de azeite. Durante o processo de extracção de azeite virgem, para o qual se recorre exclusivamente a processos físicos, produzem-se grandes quantidades de águas residuais. Estas resultam da lavagem do fruto, anterior à extracção, da água de vegetação da azeitona, e de alguma água que é adicionada para facilitar a emulsão. Estas águas residuais, vulgarmente designadas por águas ruças, tornaram-se um problema ambiental especialmente nos países da Bacia Mediterrânica. Não foi ainda estabelecido, à escala industrial, um sistema de tratamento eficiente e economicamente viável para estas águas residuais, fundamentalmente, porque são produzidas em grandes quantidades, num período curto do ano, possuem elevada carga orgânica, e apresentam teores muito elevados de compostos fenólicos e ácidos gordos. Esta tese pretende contribuir como uma abordagem de apoio ao desenvolvimento de um tratamento biológico destas águas residuais, utilizando como modelo, microrganismos adaptados do grupo das leveduras, por terem sido isolados a partir de amostras de águas ruças. Fez-se uma caracterização físico-química e microbiológica, em simultâneo, dos efluentes gerados num lagar contínuo de três fases, durante duas campanhas oleícolas consecutivas, na Cooperativa Agrícola dos Olivicultores de Murça (CAOM). A partir das amostras microbiológicas, efectuou-se um estudo de caracterização da abundância e diversidade de espécies de leveduras deste ambiente particular. As espécies identificadas em maior número de isolados foram sujeitas a um processo de selecção. Foi escolhido um isolado de Candida oleophila, cujas potencialidades para remover a toxicidade das águas ruças se revelaram mais adequadas aos objectivos. A caracterização físico-química serviu para estabelecer os parâmetros que melhor reflectiram as propriedades de cada fluxo de subproduto formado e que permitiram distinguir os tipos de efluentes gerados. Concluiu-se que as águas de lavagem (AL) são significativamente diferentes das águas da centrífuga vertical (DEC) e das águas ruças (AR). As águas de lavagem têm menor carga orgânica, pH mais elevado e teores mais elevados de oxigénio dissolvido, comparativamente com os valores registados nas águas ruças, para os mesmos parâmetros. A Carência Química de Oxigénio (CQO), Carbono Orgânico Dissolvido (COD), teores em potássio (K) e teores em fósforo (P) revelaram ser os mais adequados para caracterizar os fluxos de efluentes gerados na CAOM. A caracterização microbiológica permitiu concluir que as águas residuais de lagares de azeite apresentam uma elevada abundância de microrganismos, que incluem bactérias, fungos filamentosos e leveduras. As populações de leveduras e fungos filamentosos revelaram estar mais adaptados ao ambiente estudado, pois a sua abundância diminuiu menos quando as condições ambientais se tornaram mais desfavoráveis. Nas águas de lavagem as espécies identificadas, correspondentes a um maior número de isolados, foram a C.oleophila e Metshnikowia aff. fructicola. Nas águas ruças as espécies mais abundantes foram a C.oleophila e a C.pomicola. Por terem sido identificadas em maior número de isolados, e a partir de amostras das águas ruças, as leveduras identificadas de C.oleophila e C.pomicola foram avaliados em ensaios de degradação de compostos fenólicos. O melhor isolado desses ensaios preliminares foi seleccionado, de acordo com as capacidades registadas em utilizar vários compostos fenólicos hidroxilados e metoxilados, para destoxificar amostras de água ruça esterilizada, não diluída e não suplementada. A diminuição da toxicidade da água ruça foi verificada na germinação de sementes, na bioluminescência de bactérias, e na bioenergética mitocondrial. Concluiu-se que o isolado seleccionado de C.oleophila após um mês de incubação, promoveu uma remoção de 50% da carga orgânica das águas ruças (em termos de CQO), e 83% do teor em polifenóis totais, revelando boas capacidades depurativas. Foi ainda capaz de diminuir a toxicidade das amostras de água ruça tratada (ARt) na germinação de sementes de Lepidum sativum, aumentando o Índice de Germinação até 32% utilizando ARt diluída, comparativamente com a mesma diluição usando ARnt. Concluiu-se ainda que a ARt diminuiu a toxicidade da AR em cerca de 50% na inibição da bioluminescência em Vibrio fischerii. O teste de inibição da bioluminescência revelou ser mais sensível para avaliar a toxicidade de um efluente do que o teste de germinação. Quanto aos efeitos ao nível da bioenergética mitocondrial, concluiu-se que a acção da C.oleophila diminuiu o efeito tóxico provocado pelas águas ruças nos complexos respiratórios. Com base nos resultados obtidos, e no caso da succinato desigrogenase, o IC50 obtido mostrou uma diminuição da toxicidade superior a 50%, quando se usou a ARt. Para além do efeito tóxico sobre este complexo, verificou-se ainda um efeito deletério da água ruça sobre a ATPase. Utilizando a ARt, os valores de IC50 revelaram uma diminuição na toxicidade superior a 50%. Dadas as elevadas potencialidades demonstradas pelo isolado utilizado nos ensaios de destoxificação das águas ruças, bem como com outros tipos de efluentes, patenteou-se em coautoria, um processo de depuração de efluentes agro-industriais com elevado teor de compostos aromáticos, que tem como base a aplicação do isolado seleccionado com este trabalho de tese.
Portugal is one of the ten largest producers of olive oil. During the extraction process of virgin olive oil, exclusively done by mechanical processes, high amounts of residual waters are produced. These result from the washing of the fruit prior to extraction, from the vegetation water of the olives and some water that is added during the emulsifying process. These residual waters commonly referred to as Olive Mill Wastewaters (OMW), became an important environmental problem, especially in countries of the Mediterranean Basin. Considering industrial application of well established treatment systems, none of them has proved to be efficient and economically profitable to be applied broadly. The factors usually pointed out as responsible for that are related with high amounts of organic load, together with high concentrations of phenolic compounds and fatty acids that characterize these wastewaters. This thesis pretends to contribute with a new approach to develop biological treatment systems, using model organisms, like yeasts, isolated from samples of OMW. A simultaneous physico-chemical and microbiological characterization of OMW was done during two olive harvest campaigns, in a cooperative continuous extraction mill, of the Cooperativa Agrícola dos Olivicultores de Murça (CAOM). The results obtained in the microbiological characterization were used to study the abundance and diversity of the yeast community of this particular environment. The species identified in a higher number of isolates were used in a selection process. An isolate, identified as Candida oleophila, was selected to remove the toxicity of OMW samples. The physico-chemical characterization was used to establish the parameters that adequately reflected the wastewaters characteristics, and were able to distinguish between wastewater flows produced in the studied olive mill. It has been concluded that the washing waters (OWW) were significantly different from the waters of the vertical centrifuge (CWW) and of olive mill wastewaters (OMW). The wastewaters from OWW had lower values of organic matter, higher pH values and higher values of dissolved oxygen, comparatively to the values registered for the same parameters in OMW. Chemical Oxygen Demand (COD), Dissolved Organic Carbon (DOC), potassium (K) and phosphorus (P) values measured in the waters, have shown to be the physico-chemical parameters most adequate to characterize the residual water flows generated in the cooperative (CAOM). The microbiological characterization showed that the olive mill wastewater flows had high abundances in microrganisms, which include bacteria, filamentous fungi and yeasts. The filamentous fungi and yeast populations revealed to be more adapted to the studied environment, since their counts were less affected by the more unfavourable environmental conditions. In the OWW samples, the highest number of isolates was identified as Candida oleophila and Metshnikowia aff. fructicola species. In the OMW samples the most abundant species were Candida oleophila and Candida pomicola. Due to the fact that the species of C.oleophila and C.pomicola were identified in isolates obtained from OMW samples, and were, simultaneously, the most abundant identified species, their abilities in degrading several phenolic compounds were tested in different assays. The isolate that showed the best performance was selected to detoxify samples of sterilized, non supplemented and non diluted OMW samples. The effect of the isolate in removing some of the OMW toxicity was evaluated using germination tests, bioluminescence assays, and effects in the mitochondrial bioenergetics. The results obtained lead to the conclusion that the selected C.oleophila isolate, after one month incubation period, was able to remove about 50% of the organic matter (COD) and 83% of the total polyphenols content. The isolate was able to lower the toxicity of treated olive mill wastewater (OMWt) in the germination of Lepidum sativum seeds. The Germination Index (GI) increased up to 32% using diluted OMWt, comparatively with the same dilution using untreated olive mill wastewater (OMWnt). It was also concluded that the OMWt decreased OMW toxicity for about 50% in Vibrio fischerii bioluminescence. This inhibition test was more sensitive to evaluate the toxicity of the effluents than the germination test. As for the effects in mitochondrial bioenergetics, it was concluded that the growth of the C.oleophila isolate in OMW decreased the toxic effect in respiratory complexes. Based in the registered results, and for succinate dehydrogenase, the IC50 calculated, showed that incubation of SMP with treated decreased the toxicity more than 50%. Besides the toxic effect verified to this complex, it was also detected a deleterious effect caused by OMWnt on the ATPase. Using treated OMW, the IC50 values revealed a decrease in toxicity higher than 50%. Due to the good abilities shown by the isolate used in the detoxification assays of OMW, and also showed in other types of effluents with high concentrations of phenolics or aromatic compounds, the author of this thesis in collaboration with other inventors, patented a process for depuration of agro-industrial wastewaters with high contents of aromatic compounds, that is based in the application of the isolate selected with this work.
