Síntese, caracterização espetroscópica de novos corantes esquarílicos assimétricos e sua avaliação como potenciais fotossensibilizadores em terapia fotodinâmica

Resumo
Neste trabalho é apresentada a síntese e caracterização estrutural de dois conjuntos de
corantes esquarílicos assimétricos (Figura 1) bem como a avaliação da sua citotoxicidade in
vitro, no escuro e após irradiação a um comprimento de onda adequado, nas linhas celulares
Caco-2 e HepG2, com o objetivo de estudar o potencial dos corantes como agentes
fotossensibilizadores para aplicação em terapia fotodinâmica do cancro.
Esta dissertação encontra-se dividida em quatro capítulos. No primeiro é apresentada
uma revisão bibliográfica sobre a terapia fotodinâmica e os corantes esquarílicos. No segundo
capítulo é apresentada uma descrição de todos os métodos utilizados na síntese dos compostos
pretendidos, bem como a sua caracterização espectroscópica detalhada, e uma breve descrição
das técnicas usadas no estudo do potencial fototerapêutico dos corantes, nas linhas celulares
Caco-2 e HepG2. No terceiro capítulo são discutidos os resultados experimentais de síntese
bem como da caracterização estrutural de todos os corantes esquarílicos assimétricos. Os
compostos obtidos foram caracterizados a partir de técnicas de ponto de fusão, espectros no
vivível, espetros de ressonância magnética nuclear (RMN) de 1H e 13C e espetros de massa de
alta ou baixa resolução. Neste capítulo são também apresentados e discutidos os valores de
rendimento quântico de formação do oxigénio singuleto, obtidos para todos os corantes
esquarílicos e aminoesquarílicos a serem aplicados nas referidas linhas celulares. No quarto, e
último capítulo, são apresentados e discutidos os resultados de citotoxicidade dos corantes, no
escuro, e após irradiação com LEDs centrados a 630 ou 660 nm, durante 7 ou 14 minutos, onde
são de salientar os resultados obtidos para o corante zwiteriónico (Z = O-; Y = I) que, pode ser
considerado como um potencial fotossensibilizador para terapia fotodinâmica do cancro.
In this work it is presented the synthesis and structural characterization of two sets of asymmetric squarylium dyes (Figure 1) as well as the evaluation of their cytotoxicity, in vitro, in the dark and after irradiation at a suitable wavelength, in Caco-2 and HepG2 cell lines, in order to study the potential of these dyes as photosensitizers for use in photodynamic therapy of cancer. The present dissertation is divided into four chapters. In the first chapter, it is presented a literature review on photodynamic therapy and squarylium dyes. The second chapter presents a description of all experimental methods used for the synthesis of the organic compounds, the spectroscopic characterization as well as physical characterization and a brief description of the techniques used to study the phototherapeutic potential of the dyes, using Caco-2 and HepG2 cell lines. In the third chapter, the results of experimental synthesis as well as the structural characterization of all asymmetric squarylium dyes are discussed. The obtained compounds were fully characterized from techniques such as melting point, visible spectra, nuclear magnetic resonance spectra (NMR) of 1H and 13C and mass spectra of high or low resolution. In this chapter it is also presented and discussed the quantum yield values of singlet oxygen formation, obtained for all squarylium and aminosquarylium dyes, to be applied in cells lines referred above. In the last chapter the results of dyes cytotoxicity obtained in the dark and after irradiation with LEDs centered at 630 or 660 nm during 7 or 14 minutes, are presented and discussed; From the obtained data it is highlighted the results obtained for the zwitterionic dye (Z = O; Y = I), that can be considered as a potential photosensitizer for photodynamic therapy of cancer.
